The N-terminal half of Ypk3p localizes at the plasma membrane and is required for the activation of the G2/M checkpoint by DNA damage (Christie, et al., Science, 274, 1895-1898, 1996). In contrast, the C-terminal half of Ypk3p, which is not homologous to Hv1p, is essential for DNA damage responses. The N-terminal half of Ypk3p, which is highly homologous to Hv1p, is responsible for plasma membrane localization. The plasma membrane protein Hv1p of Saccharomyces cerevisiae belongs to a family of small tetra-span membrane proteins (Bae and Cordes, EMBO J.
